Since this is the first thread, I'll dive straight into the issue at hand.

After concluding that true happiness is only (realistically) attainable in the 2D world and with the solemn knowledge that this world does not really exist we find ourselves in the maddening state of mind we currently occupy.

The only way out beyond philosophy and resolve is to enter the world of virtual reality. Theoretically, an immersive VR world would be possible ala The Matrix albeit something thematically along the lines of the Ariaverse rather than a realistic simulation.

Now, the question is....where do we need to take science in order to make this possible. I'll be taking the biotech approach, but this will take experts in computer science, neurology, psychology, electrical engineering and many more. What will YOU do to make this happen?

Furthermore, what societal revolutions must occur such that we can relax in our virtual worlds without interruption? We must forge a world which can support this technology without public and economic backlash. As of right now, religious leaders would spurn such technology, many governments would likely treat the technology as a danger due to the level of "mind control" and brainwashing potential implied by an immersive VR tech.

Any comments thus far?

It is true that if we were to make the 2D world a lucid and concrete one, our attempts to detach from the 3D would be easier. However, with the shortcomings in technology at the present time, perhaps the only way to make up for the absence of that 2D world is to create it in our minds.
I do see anime and manga as one manifestation of the 2D realm; we may not be able to touch it, but it gives us something that we can base our own ideal 2D world upon, as well as a means through which we can distract ourselves from the temptations of the 3D world. You may ask, "but does the idea of the 2D world stem solely from the representations in anime and manga?" Perhaps. But I'd also like to believe that such views of the world had always existed in our minds, and anime and manga are what lucidified the visions that were hidden beneath the veil of 3D life on our minds, and also what brought us together here.

I don't think it'd be possible to just relax forever in this virtual world. Each person in the virtual world would be one less contributor to society and at the same time one more person relying on others for support. If everyone is rushing for the virtual world, then there will be no one left in the real world to do maintenance, etc.

Furthermore, each person will need his/her own perfect virtual world, as people themselves aren't perfect, and having another person will ruin the experience of perfection.

I think the most plausible way to go about this is to upload one's consciousness, and periodically synchronize memories. You'll have memories of experienced that perfect world, while at the same time society doesn't collapse, which means you can keep doing so. There will need to be some badass hardware though.

Great point there, NPKJI. This would be the next stage, as I see it. I think we will have realistic VR for a while before we can upload as you mentioned. VR seems to be the logical precursor technology as we'll need to collect massive amounts of data on how the mind functions in full immersion VR before we'll be able to become digital life forms.
