Obviously this anime takes place in an area of Japan where rural and urban areas intertwine as seen by short distance between the main characters' rural school and the urban Tokai no Gakuen. In episode 8, the truck driver mentions that he is driving the Tokaido Route, the southernmost road from Kyoto to Tokyo.

The original author of the manga, Neko Nekobe, was born in Hamamatsu, Shizuoka, which is along this route and also features sizable rural and urban areas.

For this reason, I think that Kingyo Chuuihou! probably takes place in Hamamatsu, Shizuoka. Well, unless anyone has any better ideas in which case I'd like to hear them. :)

It could be in Tokyo too, since the "Ina-Kanda River" from episode 37 parodies the real Kanda River in Tokyo~ it'd be disappointing tho because everywhere where that river is is pretty urban~ >_< Granted, the "Ina" part of that is supposed to refer to how it's supposed to be rural, so it's really weird still~ >_<
